More than 100 councillors from the larger Gusii on Sunday declared they would back the 'Yes' campaigns irrespective of their party affiliations.
The leaders arrived at the decision after a meeting chaired by Public Works minister Chris Obure at a Kisii Hotel.
Speaking after the meeting, Mr Obure said they would join hands to persuade people in the region to back the proposed constitution in next month's referendum.
Mr Obure was optimistic that the Green team would carry the day. "We can't afford to lose such a good constitution. It is what we have been searching for for two decades," he said.
"Kenyans need to pass the new constitution because it is good for them and the generations to come. They should not listen to cheap propaganda from the 'No' team, which is capitalising on a few clauses to ensure that the whole document is rejected."
The minister, who was flanked by chairmen from the counties in the area, refuted claims that there had been differences between him and the minister for Education Prof Sam Ongeri, on who was to steer the 'Yes' campaigns in the region.
Common stand
Mr Obure insisted that they had a common stand on the proposed constitution as leaders from the area. "We have resolved as elected leaders from this community to spearhead the 'Yes' campaigns together," he added.
The minister said that they would embark on vigorous campaigns in the entire region on Tuesday. Kisii mayor Samuel Nyangeso, who is steering the 'No' campaigns in the region, did not attend the meeting.
